Abstract
Human papillomavirus (HPV) could be detected by various molecular biological methods. We compared the detection rates and advantages of various methods such as Southern blot hybridization. In situ hybridization, in situ polymerase chain reaction
(PCR),
PCR and PCR-SSCP (single stranded conformation polymorphism). Although in situ hybridization have relatively lower sensitivity (82.3%), this method could detect the localized HPV DNA in the tissue. Southern blot hybridization could detect HPV in
89.3%
of condyloma acuminata and have the advantage of identification of subtype of HPV. The method using PCR shows higher sensitivity (100%) but PCR needs various set of primers for detection and tvping of HPV from HPV infected clinical lesions. A
newly
developed method, PCR-SSCP is found to be very simple and rapid for the detect of HPV. Instead of multiple sets of primers, this method enables us to reduce the number of PCR experiments for the detection of HPV by using common primers for HPV.
